Palm-Glu(OSu)-OtBu (CAS 204521-63-1), also known as N-palmitoyl-glutamic acid alpha-(tert-butyl ester) gamma-(N-hydroxysuccinimide ester), is an activated NHS ester form of the palmitoyl-glutamic acid building block, of the fatty acid-activated ester-amino acid chemical class. The N-hydroxysuccinimide (OSu) active ester at the gamma-carboxyl allows for controlled reactivity, which makes it possible to selectively form amide bonds with primary amines (lysine ε-NH2) in mild aqueous or DMF conditions. Process impurities from incomplete activation include the free acid (Palm-Glu-OtBu) and the palmitic acid NHS ester by-product.
As a pharmaceutical-grade activated SPPS reagent for GLP-1 fatty acid acylation chemistry and quality control, ApiSyn sells Palm-Glu(OSu)-OtBu (CAS 204521-63-1).
