Liraglutide (CAS 204656-20-2) is a 31-amino acid GLP-1 receptor agonist peptide with a C16 fatty acid (palmitic acid) linked to Lys26 via a glutamic acid-mini-PEG linker. It belongs to the fatty acid-acylated GLP-1 analogue chemical class. The sequence is His-Aib-Glu-Gly-Thr-Phe-Thr-Ser-Asp-Val-Ser-Ser-Tyr-Leu-Glu-Gly-Gln-Ala-Ala-Lys(N-ε-palmitoyl-γGlu-OEG)-Glu-Phe-Ile-Ala-Trp-Leu-Val-Arg-Gly-Arg-OH. FDA approved it for type 2 diabetes (2010), obesity (2014, as Saxenda), and lowering the risk of heart disease (LEADER trial, NEJM 2016) in people with diabetes who already have heart disease. The palmitoyl-γGlu lipid modification allows albumin to bind, which makes the drug last about 13 hours and allows for once-a-day injections. The main building block is Fmoc-Lys(palmitoyl-Glu-OtBu)-OH (CAS 1491158-62-3). Deamidation at Gln28/Asn and oxidation at Met/Trp are examples of process impurities. There are also deletion peptides, lipid-unconjugated Lys26 impurities, and fatty acid chain-length analogues.
