Fmoc-Ser(tBu)-OH (CAS 71989-33-8), also known as Nα-Fmoc-O-tert-butyl-L-serine, is the standard doubly protected serine for Fmoc-SPPS, of the Fmoc-serine tBu ether class. Serine is found in many peptide positions that are important for therapy, such as GLP-1 analogues (Ser8 in native GLP-1, which is essential for GLP-1R binding), GnRH (Ser4), Teriparatide (multiple Ser), Vasopressin, and Oxytocin. Under basic conditions, serine undergoes β-elimination to yield dehydroalanine and is susceptible to O-acylation side reactions in Fmoc-SPPS.
