Fmoc-Pro-OH (CAS 71989-31-6), also known as Nα-Fmoc-L-proline, is the standard Fmoc-protected proline for Fmoc-SPPS. It is a member of the Fmoc-secondary cyclic amino acid class. Proline is a secondary amine that adds special features to peptide backbones, such as cis/trans prolyl peptide bond isomerism (which is crucial in peptide biology), β-turn nucleation, and protease resistance (most endoproteases take a long time to break Pro-X bonds). Proline is found in many therapeutic peptides, such as GnRH (Pro9), Leuprorelin , Buserelin, and many hormone mimetics.
