Fmoc-Phe-OH (CAS 35661-40-6), also known as Nα-Fmoc-L-phenylalanine, is the standard Fmoc-protected phenylalanine for Fmoc-SPPS. It is an aromatic amino acid. There is no need to protect the side chain of phenylalanine; its benzyl side chain is stable under both Fmoc-SPPS (piperidine) and global TFA deprotection. Phe is found in many therapeutic peptides, such as GLP-1 analogues (Phe6, Phe28 in native GLP-1/semaglutide), octreotide (D-Phe1, Phe3), GnRH (His2-Trp3-Ser4-Tyr5-Gly6-Leu7-Arg8-Pro9 with Phe not in standard GnRH, but in analogues), vasopressin, and antimicrobial peptides.
