Fmoc-Lys(ipr,Boc)-OH (CAS 201003-48-7), also known as Nα-Fmoc-Nε-isopropyl-Nε-Boc-L-lysine or Fmoc-Lys(iPr-Boc)-OH, is a specialty Fmoc-SPPS building block with an N-isopropyl-Boc (iPr-Boc) protecting group on the lysine ε-amino group. N-alkylated lysine residues can change how well H-bonding works, add N-methylation-type conformational constraints, and make the final peptide less likely to break down by proteolysis. N-isopropyl-lysine residues are found in metallopeptide chelators that were made on purpose, peptide libraries that are limited, and special peptidomimetics. Piperidine usually takes off the Fmoc group, while extended TFA treatment or HF deprotection takes off the Boc group from the sterically encumbered secondary amine.
