Fmoc-Lys(Boc)-OH (CAS 71989-26-9), also known as Nα-Fmoc-Nε-Boc-L-lysine, is the standard doubly protected lysine for Fmoc-SPPS. It belongs to the Fmoc-lysine Boc epsilon-amino class. This is one of the most common Fmoc amino acids used to make pharmaceutical peptides. Boc protects the ε-amino group (TFA takes it off in global deprotection), and piperidine takes off the α-Fmoc at each SPPS cycle. Lysine is the acylation site in GLP-1 fatty acid-modified peptides (Liraglutide Lys26, Semaglutide Lys34, Tirzepatide Lys26, Retatrutide Lys24). After global TFA deprotection, the free ε-NH2 can be used to couple with a fatty acid side chain or make other changes.
