Fmoc-Leu-OH (CAS 35661-60-0), also known as Nα-Fmoc-L-leucine, is the standard Fmoc-protected leucine for Fmoc-SPPS. It belongs to the Fmoc-branched aliphatic amino acid class. Fmoc-Leu-OH is one of the most popular SPPS building blocks because leucine's isobutyl side chain doesn't need any extra protection. Found in GnRH analogues (Leu7 in native GnRH), Leuprorelin (Leu7), Octreotide (Leu4 equivalent), vasopressin analogues, and many other therapeutic peptides. Leu is a hydrophobic amino acid that plays a big role in making peptide helices stable and forming hydrophobic cores. Coupling Fmoc-Leu-OH usually works well, but in long hydrophobic sequences (poly-Leu), β-sheet aggregation on resin can make it less effective.
