Fmoc-Ile-OH (CAS 71989-23-6), also known as Nα-Fmoc-L-isoleucine, is the standard Fmoc-protected isoleucine for Fmoc-SPPS. It belongs to the Fmoc-branched aliphatic amino acid class. Isoleucine has two stereocenters, Cα and Cβ, and both must be kept the same during synthesis. Ile is found in somatostatin analogues, tirzepatide, and a variety of pharmaceutical peptides. Being a β-branched amino acid, coupling Fmoc-Ile-OH can be hard on the sterics. In some cases, longer coupling times or double coupling are needed.
