Fmoc-His(Trt)-OH (CAS 109425-51-6), also known as Nα-Fmoc-N(im)-trityl-L-histidine, is the standard doubly protected histidine for Fmoc-SPPS. It belongs to the Fmoc-histidine trityl imidazole class. The trityl (Trt) group protects histidine's imidazole side chain (N(τ)) from alkylation and oxidation side reactions during Fmoc-SPPS and lowers the chance of racemization during α-amino acid activation. In the last step of global deprotection, TFA/scavengers take away the Trt group. His is very important at the N-terminus of GLP-1 (His7 in native GLP-1 and His1 in the semaglutide/liraglutide sequences, which are very important for GLP-1R binding), in GnRH (His2), and in many peptide hormones.
