Fmoc-Gly-OH (CAS 29022-11-5), also known as N-(9-fluorenylmethyloxycarbonyl)glycine or Nα-Fmoc-glycine, is the simplest and most commonly used Fmoc-SPPS amino acid building block. It belongs to the Fmoc-amino acid class. Glycine is achiral (no side chain stereocentre), doesn't need any protection for its side chain, and is found in almost all therapeutic peptides. It can be found in GLP-1 sequences (Semaglutide, Liraglutide), oxytocin, vasopressin, GnRH, and a lot of other peptide drugs. Fmoc-Gly-OH is the standard building block for developing the Fmoc-SPPS method, testing coupling efficiency, and improving resin loading. Fmoc-Gly-OH is also used as a standard for loading in SPPS resin loading tests and as a calibrant for HPLC gradient calibration.
