Fmoc-Gln(Trt)-OH (CAS 132327-80-1), also known as Nα-Fmoc-Nε-trityl-L-glutamine, is the standard doubly protected glutamine for Fmoc-SPPS, which is part of the Fmoc-glutamine trityl amide class. To stop glutamine's main amide (γ-CONH2) side chain from dehydrating to glutaminyl nitrile during carboxyl activation and to stop it from cyclizing to make pyroglutamate (pGlu) from N-terminal Gln in peptide sequences, it needs to be protected by Trt. TFA/TIS (triisopropylsilane scavenger) takes away the Trt group.
ApiSyn sells Fmoc-Gln(Trt)-OH (CAS 132327-80-1) as a pharmaceutical-grade Fmoc amino acid and a key raw material reference standard for GMP peptide SPPS and deamidation impurity analysis.
