Fmoc-D-(TIC)-OH (CAS 130309-33-0) is a Fmoc-protected, conformationally constrained, unnatural D-amino acid of the tetrahydroisoquinoline-3-carboxylic acid (Tic) class. It is also known as Nα-Fmoc-D-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid. It has rigid psi angle constraints that mimic a β-turn conformation and make it resistant to enzymatic proteolysis. GnRH antagonists like Cetrorelix, Degarelix, and Ozarelix use the D-configuration (D-Tic) to lock in the specific backbone dihedral angles needed for GnRH receptor antagonism. Fmoc-D-Tic-OH is an important part of making Degarelix, an injectable GnRH antagonist for prostate cancer. The enantiomer is Fmoc-L-Tic-OH (CAS 130309-37-4).
