Desmopressin Acetate (CAS 62288-83-9) is a synthetic version of the neurohypophyseal hormone arginine vasopressin (AVP). It is a cyclic disulfide vasopressin nonapeptide analogue. Desmopressin is [1-(3-mercaptopropionic acid)-8-D-arginine]-vasopressin, which means that two parts of its structure have been changed: the N-terminal amino group has been removed (deamino-Cys1 → β-mercaptopropionic acid) and the D-Arg8 has been replaced. These changes make the plasma half-life longer (about 6 to 24 hours instead of 20 minutes for AVP) and make the drug more selective for V2 (renal antidiuretic) receptors than V1a (vasopressor) receptors. Approved for von Willebrand disease, central diabetes insipidus, primary nocturnal enuresis, and bleeding in hemophilia A. Some of the most important impurities are linear desmopressin (non-cyclized), desamino-deamino (over-reduced species), D-Arg→L-Arg epimer, and Gln4→Glu4 deamidation product.
