Cbz-Thr(tBu)-OH (CAS 16966-09-9), known as N-benzyloxycarbonyl-O-tert-butyl-L-threonine or Z-Thr(tBu)-OH, is a Cbz/Z-protected threonine with O-tBu side chain protection, belonging to the Cbz-threonine tBu ether amino acid chemical class. This compound is mainly used in classical peptide chemistry, which is the synthesis of peptides in solution, and in hybrid SPPS/solution routes, where the Cbz group protects Boc or Fmoc protecting strategies in a different way. Under mild conditions, hydrogenolysis (H2/Pd-C) takes away the Cbz group, and TFA takes away the tBu ether on the threonine hydroxyl. Epimerization at threonine's Cα is a concern in both solution and SPPS.
