Amino Acids Ac-D-2-Nal-OH 37440-01-0 Ac-D-2-Nal-OH (CAS 37440-01-0), also known as N-acetyl-D-2-naphthylalanine or Nα-acetyl-(R)-3-(naphthalen-2-yl)alanine, is a N-acetyl-protected unnatural D-amino acid of the naphthalene-2-yl-D-alanine (D-Nal) chemical class. This compound is a specialty SPPS building block that is mostly used to make GnRH analogues (decapeptide hormone agonists and antagonists). The D-2-Nal residue at position 6 is very important because it keeps the molecule from changing shape and makes it resistant to proteases. The big, aromatic naphthalene side chain interacts with the GnRH receptor binding pocket through π-π stacking and hydrophobic interactions. The acetyl N-protecting group is not as common in SPPS as Boc or Fmoc, but Ac-D-2-Nal-OH could be used as a reference standard for amino acid analysis, checking for racemization control during synthesis, and figuring out how D-Nal residue-specific enzymatic hydrolysis works. It is very important to have enantiomeric purity because the L-Nal impurity would be a racemization product during synthesis.
