(2S)-2-[[4-(Trifluoromethyl)phenoxy]methyl]oxirane (CAS 256372-58-4) is a chiral aryloxy-methyl epoxide of the (S)-trifluoromethylphenoxymethyl glycidyl ether chemical class. This enantiopure epoxide is a key building block for making many drug candidates and APIs that are sold. The (2S)-glycidyl-CF3-phenyl ether is used to make antifungal agents (like itraconazole and posaconazole) and antiallergic scaffolds, such as intermediates related to Rupatadine. The highly electrophilic, strained oxirane ring undergoes regioselective SN2 ring-opening by nitrogen nucleophiles (amines, triazoles) to provide chiral amino-alcohols or azole-propanol fragments essential in azole antifungal pharmacophores. The trifluoromethyl group makes the molecule more stable and lipophilic (Hansch π +0.88). Maintaining chiral fidelity in downstream APIs depends on controlling enantiomeric purity (ee ≥99%).
